Histological Investigation of the Appetite Center in Antler-administered Mice


Abstract
Time-honored herb tonic, antler has been widely administered on the thin patient in this country despite of its expensiveness. Traditionally people believes that antler has continuous tonic effect even far later date after the administration of its water extract.
In 1975, author observed irreversible tissue change of inhibitory center of the appetite resulting voracious appetite and weight-gain in gold-thiogluse-injected mice. Accordingly this study was performed to investigate whether water extract of antler induced histological change on such center or not assuming irreversible tissue change might explain continuous tonic effect.
Materials and Method: A total of 60 male albino mice were fed with commercial rodent feed; 20 out of 60 were fed contrast group, 20 were fed adding 150mg/ kg/ day of the water extract of antler for 4 weeks and the rest 20, 300mg/kg/day respectively. Thereafter all of groups were fed with rodent feed not containing antler extract for further 1 -8 weeks until being sacrificed.
Results: A group of rodent fed with 300mg/kg/day of antler extract seemed to show weight-gain in comparison with the other groups. The difference of body weight, however, was variable even in same group. There were no statistical signifiance. Microscopically there was also no significant histological change between each group although same questionable increase or decrease of cellularity and/or degree of staining of hypothalamic cell groups were observed on both feeding and satiety centers.
Conclusion: The significant histological change did not develop on the appetite center with administration of the water extract of antler for 4 weeks in the mice.
